For some backstory, we showed some time ago that inactivating ALL mOFC outputs increased pursuit of larger, risky rewards, even if this wasn’t always the best play- similar to findings in humans.

Now, the mOFC is reciprocally connected with both the basolateral amygdala (BLA) and prelimbic regions of the anterior cingulate PFC, and both of these regions also refine these types of decisions. We asked, how do these regions talk to each other and what type of info do they process.

Using a chemogenetic approach, we found that disrupting top-down BLA➡️mOFC circuits made animals inflexible in their decisions- suggesting this circuit tracks changes in relative value to support flexible reward-seeking

In comparison, when we suppressed Prelimbic inputs to the mOFC, that made rats more risky, implying that this cortico-cortical circuit attenuates the allure of larger yet uncertain rewards and promotes loss sensitivity

What was really cool was when we disrupted mOFC-to-prelimbic inputs. This induced more random choice patterns, so animals were less risky when the odds were good, and more risky when they were poor
