Recently, Conor McFadden in @retof.bsky.social's group introduced a neat data processing trick that exploits aliasing to speed up oblique plane microscopy more than two-fold, without the need to change any hardware.

In the original work, aliased copies of the Fourier spectrum were moved to their correct positions in Fourier space before a fully sampled spectrum was cropped out. Here, we show how the problem can be attacked using deconvolution, producing denoised and deconvolved volumes as a by-product.

Next, we apply the algorithm to high-resolution OPM data, looking at diffraction-limited clathrin puncta in a single cell. As expected, the algorithm outperforms bicubic interpolation and, most importantly, correctly fails when the undersampling factor is too large.

Then, we use our new direct-view oblique plane microscope (https://doi.org/10.1364/OPTICA.558420) to image a 4× expanded brain organoid with 5-fold undersampling. While the new algorithm is slower than the previous approach, it avoids artefacts and produces denoised or deconvolved volumes simultaneously.

We hope that aliased acquisition becomes the standard approach for oblique plane microscopy, as it significantly increases acquisition speed with no penalty in spatial resolution. With our new algorithm, we can produce a fully sampled deconvolved volume faster than in a normal acquisition.
