The technique combines the application of an external electric field with a sensitive readout of fluorescence lifetime changes caused by molecule redistribution inside a nanocavity. Our approach is calibration-free, experimentally (relatively) simple, and compatible with single-molecule resolution.
We demonstrate that the method accurately detects the sign and magnitude of molecular charges and that it can also be used for determining the ionic strength of the surrounding solution. We develop a through theoretical framework of the idea and present proof-of-principle experiments.
We hope that our method opens new avenues for precise, real-time measurements of biomolecular charges — a fundamental parameter in understanding biochemical interactions and molecular function in biology.
