First off my conflict of interest: I am a long established paid consultant for Oxford Nanopore, been advising them since 2011, and a shareholder. No company (ONT included) vets what I say.

On the "vanilla" side of DNA sequencing, there is now steady progress on quality and automation, and the length of ONT reads has always been amazing. I expect we will see more updates both in the single molecule, single base error rate and the consensus quality.

It's been clear for 12-18months that ONT reads alone make very accurate and complete bacterial genomes - marching this up to human is the thing. The most important parts of human - exons (coding or non coding) thankful exclude the most annoying sequencing areas (homopolymers) >>

<< this means the practical quality for (say) rare disease and cancer genomes in humans is there now, and it is just going to get better.

(A long standing issue is where the single molecule quality tops out, but I think we're still a long way off true technical limitations. Much of this is now a deep learning problem and basically - faster, bigger NVIDIA chips, more accurate calls, and NVIDIA are definitely making faster bigger chips)

@lh3lh3.bsky.social's latest assembler has really pushed using "routine" ONT libraries (N50s 30KB ish) to get to near T2T assemblies, leveraging read length for (diploid) error correction. Heng continues to be the fountain of excellent sequence bioinformatics tools and it impressive to see.
