Neuroendocrine (NE) cancers often have tuft-like cells, not yet modeled in the #lung. We show with two methods in multiple mouse models that targeting genetic changes found in tuft-like SCLC (high MYC, low ASCL1, Pten loss) leads to #POU2F3 SCLC, from a basal cell, not NE cell.

Basal cells are more proliferative and abundant than NE and tuft cells. Basal cells respond to injury by making NE and tuft and are found in locations of human sclc.

#Ionocytes are another rare basal derivative and we find their signatures in sclc.
Prior work shows adenoa go thru a basal-like state on their way to NE.
Our new single cell lineage tracing in organoid models support basal plasticity to all of these states.

Often expresses YAP1 and has a Tcell-inflamed signature. We show YAP1 is expressed in basal cells and also has inflamed signature. Using the largest real world SCLC dataset to date, we show the mouse remarkably resembles human from a basal cell and that an inflammatory basal state exists in human
