To better answer this question, we first built a new reporter system based on genomic integration of the R-loop forming mAIRN sequence. We show that your system can efficiently induce TRCs, R-loops, and DNA damage. (2/n)

Focusing on chromatin, we show that TRC induction stabilizes R-loops, which causes a drastic reduction of local nucleosome occupancy, suggesting that R-loops are incompatible with nucleosome formation. (4/n)

Intrigued by our findings on nucleosome dynamics, we screened several histone modifications for an association with TRCs. Strikingly, we discover H3K79me2/3 as a TRC-associated histone modification. (5/n)
