TFIID is a large multiprotein complex known for its functions in transcription initiation. Like many other protein complexes involved in gene regulation, TFIID contains intrinsically disordered regions (IDRs). Do these IDRs have a function?

We identified an IDR in the TFIID subunit TAF2 that partitions TAF2 into nuclear speckles. These condensates contain proteins involved in RNA splicing, RNA processing, and RNA export. Surprisingly, TAF2 is the only TFIID subunit, which accumulates in speckles!

What’s special about the TAF2 IDR? It contains an evolutionarily conserved stretch of histidines and lysines, which drives TAF2 IDR phase separation in vitro and targets TAF2 to nuclear speckles in cells. This sequence is partly even conserved in plants!

We identified 3 nuclear TAF2 protein pools: 1) TAF2 as part of a nucleoplasmic TFIID complex, which is not associated with chromatin. 2) TAF2 in the outer shell of nuclear speckles where it interacts with the splicing factor SRRM2. 3) TAF2 localized to pol II promoters as part of the TFIID complex.

Why is TAF2 sequestered in nuclear speckles? We performed TAF2 proximity labeling and found that it interacts with splicing factors, such as members of U2 and U5-type snRNP complexes. The TAF2 IDR seems to modulate TAF2’s interaction frequencies with RNA-binding proteins.

Does TAF2 have multiple roles in the regulation of gene expression? Yes. 1) TAF2 regulates promoters: Deletion of the TAF2 IDR leads to increased TAF2 promoter association. 2) TAF2 influences RNA splicing: Modulation of TAF2 protein levels and IDR deletion elicit alternative splicing events.
