This protein is present in diplonemids/bodonids/euglenids/Naegleria, which have two flagella, but not in trypanosomatids that have just one flagellum. Shedding has not been reported in this organism, to my knowledge. The length seems to change during cell cycle though. I like that fact that so far I can find this gene only in biflagellate, not monoflagellate

Is it the anterior or the posterior flagellum? And, which euglenids did you look in?

Diplonema papillatum, which is a sister of euglenids. As far as I know, these two flagella have not been defined as anterior vs posterior (both come out of anterior end of the cell). This picture is from Wikipedia. EM studies say these two look nearly identical https://en.wikipedia.org/wiki/Diplonemidae

Thanks! So it's apparently in diplonemids/bodonids/Naegleria and there's no data from euglenids (though it was presumably in their shared ancestor with diplonemids), is that right?

The protein is present in euglenids as well as I mentioned above. So there is correlation between its presence and being biflagellate

In other diplonemids, the anterior/dorsal flagellum has been defined as the one forming loop around the anterior of the cell, with the posterior/ventral one trailing behind in a posterior direction (see Tashyreva &al https://doi.org/10.1128/mbio.01921-23, Montegut-Felkner& Triemer 1996)

If yes, you may check whether it was by chance stained in ciliated human cells in the human protein atlas and whether you see diversity between different cells: https://www.proteinatlas.org/. Or, if the protein exists in humans, you could check whether the human protein atlas contains antibody stainings for the protein on human tissues with motile cilia (https://www.proteinatlas.org/humanproteome/tissue) and whether you can observe a similar phenomenon there as well.

He was my undergrad mentor and they worked a lot of tubulins and specifically, as I recall, gamma tubulin localized to the basal body. I haven't been in touch in some years. You may also know about Dr. Lillian Fritz Laylin and a researcher who used to post a lot on X, Dr. Katarina Velle.
