Let me first tell you how we started this project. Although we had already looked carefully at sorted worm neurons previously via RNAseq– in wild-type, daf-2, and with age (cite Weng) – we had not used snSeq to address, what are the transcriptional changes in specific neurons of daf-2 worms that allow them to have better learning and memory.

To ask this question in a mutant requires that you carry out the experiment in that mutant, you can’t rely on a database for that. Moreover, the existing database the field uses (CeNGEN) is excellent but are not from adults, they are late larval (L4 animals) stage.

The problem is, L4 animals can’t carry out our standard associative learning, so we also needed to figure out what changes between L4 and adulthood.

But previous methods for snSeq were quite laborious. Instead, we modified what we had done in mice and combined it with our methods for adult neuron isolation, and collected wild-type and daf-2 neurons.
