Interesting, but makes sense. Each organism's DNA can vary in size, so there isn't a mechanism to guarantee a specific amount is needed/required.

In a single hypotriploid cell line generated through adenoviral transformation of embryonic kidney cells. Interesting maybe, but not clear how generalizable to cells other than HEK293, which are atypical in many aspects.

They certainly do a fair bit of loss of function characterization across major DNA maintenance proteins, so it sets up a system to test in other cells. But I think it’s a bit of a stretch to conclude that their results challenge the paradigmatic existence of a G2/M checkpoint…

I don't think they claim there's no G2/ M checkpoint at all. The way I understand it - the cells sense the breaks resulting from underreplication and stop. But once the breaks are repaired, they go into mitosis and don't care that the replication is not completed. Am I missing something?

Your point’s well taken. They rename the G2/M checkpoint as a “replication completion checkpoint” and conclude it doesn’t exist because underreplicated HEK293 cells can progress to mitosis after DNA repair. Probably true in HEK, but they’ve already broken all sorts of genome maintenance rules!

I do agree with you that it would be very cool to see if this system exists in other cells. Could be an interesting mechanism of tissue evolution in developing organisms, for instance.
