My lab stored poplar seeds in LN2 for 10 years without a decline in germination rates. Poplar seeds are notoriously difficult to store.

Yet most genebanks do not use liquid nitrogen to conserve seeds. Different situation for vegetative species-cryopreservation. -18C is fine for long term storage.

A way to shock stratify Setaria seeds that was common in the Kellogg lab was just to chuck them in the -80C freezer for 48-72 hours. Generally still prefer stratifying this way over chemicals since it’s faster and relatively more consistent.

Not really but they did it on 24 species so they must've had a hell of a system
