Beyond the ordinary repertoire of cell biological and biochemical approaches, we use various imaging techniques (incl. EM+CLEM) and proximity proteomics (our favourite tool is the APEX enzyme). We have also established zebrafish models in the lab, which I'm very excited about.

Some recent highlights from the lab: 1) In a recent preprint we have identified a molecule (ARHGAP12) that controls the junctional actin cytoskeleton to regulate the paracellular flux of small macromolecules across tight junctions. https://doi.org/10.1101/2024.08.24.609485

Our data indicate that ARHGAP12 suppresses the activity of N-WASP to dampen actin assembly via Arp2/3 complex. This limits junctional tension and makes tight junctions more leaky. The story is currently under revision. Thanks to the two reviewers who have been very kind to us!

2) Using time-resolved proximity proteomics with APEX2 we have identified a membrane tension-sensitive caveolin-1 interactome that is likely to be important for the function of caveolae at the rear of migrating cells. https://elifesciences.org/articles/85601.

3) we have interesting new data on the functions of the Homer proteins in epithelial cells (which we recently identified to interact with the Crumbs complex), and we have news on how apical proteins are trafficked during epithelial lumen formation. All unpublished - stay tuned!
