Mammalian nuclei are incredibly dense environments, with nearly two meters of DNA jammed into a space only 10 microns wide. Cells overcome this compaction issue architecturally—by sequestering most genomic processes into functionally distinct compartments. (3/30)

Over the past few decades, our field has developed many powerful tools for probing this compartmental architecture. But there are still core aspects of nuclear organization that remain almost impossible to interrogate by existing technologies. (4/30)

Notably, few tools are available for the targeted biochemical characterization of single loci. If you want to dissect the "neighborhood" around an individual locus—unbiasedly discovering the nearby chromatin domains, RNAs, and proteins—you're in for a challenging time. (5/30)

Just think about all the microcompartments in a nucleus—speckles, enhancer hubs, transcription condensates, chromocenters, repair foci, etc. Are all compartments of a given type the same? Or do individual compartments adapt to the specific needs of the loci they touch? (6/30)

Congratulations on these multiple achievements!! 😍 is it too ambitious to adapt it, as a follow up, to identify neighbouring lipids? This would be an even greater revolution. By including lipids in the nuclear picture, a further dimension will unfold! @morielab.bsky.social will be eager to know!!
