Indeed such a striking difference is really what puzzles me. Maybe I miss something obvious in the acquisition, but maybe not. At least now I have plenty of interesting answers to investigate!

And it goes exactly at the opposite of what I observe: the molecule is Tag-RFP-T: it is supposed to be the most photostable one, both in WF and in LSCM. This needs more investigation time that I can afford.
