First of all, we recommend reading a complementary study led by Yannick Baffert and @sbigot.bsky.social, in which they use Tn-seq to study the genetic determinants of pOXA-48 in plasmid maintenance and conjugation. Really cool piece of work that reaches similar conclusions to ours!

We know that pOXA-48 leads to very different fitness effects in its several bacterial hosts. The question is, what are the genetic basis behind said differences? We perform CRISPRi screens in different clinical bacteria to study the role of each plasmid gene in fitness cost and plasmid biology. 3/14

Because pOXA-48 leads to such host-specific fitness effects, we expected the response to individually silence each plasmid gene to be strongly host-specific as well. Surprisingly, that is not the case. 4/14

The genes encoded in the plasmid produce similar fitness effects across the different enterobacterial hosts. There are small differences between strains, but we observe a clear general trend across them. 5/14

We identify the genes important for pOXA-48 across the different strains. Some of them are involved in plasmid maintenance and mitigation of plasmid-cost by modulation of gene expression. Individually silencing of these genes leads to plasmid-loss at different rates in the several strains. 6/14

We also show that the PemI-PemK toxin-antitoxin system promotes vertical transmission of pOXA-48 to cell progeny. However, the essentiality of the system for plasmid maintenance in the population varies depending on the species used. 7/14
