What do you think would win, stiffness or ECM gradient? Could you counter balance one over the other?

We have not done the experiment, but I bet ECM ligand density wins over stiffness, at least for single cells. Single cell durotaxis is very inefficient. Also, haptotaxis is just about passive friction differentials, whereas n durotaxis you need myosin to pull on the surroundings. For cell groups… 🤔

One of my dream experiments would have been a cell gym kana... It is technically feasible, e. g., with Primo PA and SU8 and protein patterning. Digital patterns might be of interest as well... I'm getting enthusiastic here...

We show that cells exposed to micropatterned fibronectin gradients exhibit a complex repertoire of trajectories, including directed haptotactic migration up the gradient but also linear oscillations and circles with extended periods of migration down the gradient.

Using a coarse-grained molecular clutch model, we show that the observed complex trajectories over long time scales arise from the interplay between differential friction, persistence, and physical confinement.

as long the cells don't eat the coating it is cool. Quite surprisingly, to me, my guess would have been the cells stop in an area of high fibronectin density

Exactly! This was also one of our predictions based on existing clutch models. But strong polarity persitence allows the overshooting. This is why we developped a clutch model with polarity persistence.
