This study builds on our previous work on the evolution of gene expression and chromatin accessibility across the development of the mammalian cerebellum (https://doi.org/10.1038/s41586-023-06884-x, https://doi.org/10.1126/science.abg4696), a brain region increasingly implicated in the evolution of human cognition.

We generated new single-nucleus chromatin accessibility/multiome datasets for human (10 stages), bonobo (2), macaque (1) and marmoset (8). By integrating these with our published data for human, mouse and opossum, we created a resource spanning 780,000 single-cell profiles across six species.

Using our uniform annotation of cell types and states across species, we discovered that mouse-trained models perform nearly as well on human sequences and vice versa, revealing conserved enhancer grammar in mammalian cerebellar cell types.
