Summarize so that what might help to market the book is emphasiszed. Mention the unique setting, maybe the attention to historical detail or the colorful characters -- anything that makes the book stand out. Mention your expertise in some aspect of the book, maybe a colorful relevant detail.

It's largely in the details mentioned in the query. In my case, for instance, mentioning the Pacific Northwest was a setting is seen as a major selling point by interested agents. The mistake many make is to summarize but not chose a slant on it.

The best query letters also include the book's marketing potential, which pretty heavy lifting in itself.
