In summary, our study represents the largest meta-analysis of gut microbiome associations with CRC.
This study was led by Gianmarco Piccinno and we would like to thank @cibiocm.bsky.social members, the ONCOBIOME Consortium and the PROSPECT team for this great collaboration.

We also investigated the within species differential gene carriage and strain-differentiation between controls and CRC.

We studied the gut microbiome composition along stages of CRC and primary tumor location.

We exploited machine learning and meta-analysis approaches to characterize the association of the gut microbes with CRC.
The gut microbiome signature is characterized by peculiar species which are very unlikely inhabitants of the healthy gut!

We leveraged >3,700 fecal samples derived from healthy, adenoma and CRC individuals and sequenced with shotgun metagenomics.

๐1,816 samples (19.6%) were positive for human mtDNA (see Fig 1A)
๐Positive samples spanned over 21 countries, w/ China, Sweden, Canada, Israel, and Mongolia having the most positives (see Fig 1B)
๐Positive cases were derived from stool, oral & skin samples (see Fig 1C)

We conducted an exploratory analysis of 9,251 publicly available human metagenomes from 31 countries & 48 cohorts that we previously published,
and discovered a significant presence of human mtDNA sequences
plus investigated the extent of mtDNA presence.

In short:
๐ We analyzed 32,152 metagenomes from 94 studies across the ๐ to investigate the human microbiome intraspecies genetic diversity.
๐ We reconstructed 583 species-specific phylogenies and linked them to geographic information and speciesโ horizontal transmissibility.
