How is this different from sequencing 96 bacterial isolates ? Do you mean you're also paying for them to do something with the 96wells?

Are you asking a per genome cost? We usually budget ~$85 AUD per genome for illumina, but that doesn’t include labor because we seq in house. You can do it cheaper if you 1/4 the library prep reagents

Thanks! I guess I'm just surprised at how much it costs to sequence little genomes. But I think your message points out where I've been thinking about it wrong. Human time and library prep costs are fixed, so even though sequencing is super cheap for small genomes, the price remains high.

No worries! Once you 1/4 reagents the library prep cost comes down a lot. It’s the DNA extraction that’s then the big chunk. My estimate btw is for 5Mbp, so 3.5 should be a fraction cheaper because you can fit a few more on a flow cell

Exactly. Our illumina provider offers twice as much data for 10USD ekstra per genome, but base price remains around/below 100USD per bacterial genome.

@roblanfear.bsky.social short or long reads? We should be able to give you ~200x ONT SUP for $60 a genome or less, I closing sample QC. I suspect short reads will be significantly cheaper.
