For Chelsea, it's because they played away in Kazakhstan on Thursday night, and arrived back in the UK on Friday morning. Not sure about Southampton Spurs though.

Mmm…Chelsea played their Conference League team as apposed their EPL team resting in London.
Spurs played away same night, same rules apply except for the non-availability of injured/suspended players.

The difference is that Spurs were only playing in Glasgow, one of the easiest European away days. If you applied that to them then the rule would have to apply for any team that played away anywhere on Thursday.
