Our method provides rapid access to chemically modified lasso peptides and enables the investigation of SAR, epitope grafting, and the improvement of therapeutic properties.

While precursor peptide synthesis worked well (57 AA), the recombinant expression of the maturation enzymes McjB & McjC required extensive optimization. Once optimized, both enzymes showed a remarkable promiscuity tolerating almost all non-natural modifications!
